Cellular Kinase Assays

Alpha SureFire® assays are no-wash cellular kinase assays. While they exist in different formats (AlphaScreen® SureFire, AlphaLISA® SureFire Ultra, and Alpha SureFire Ultra Multiplex), all use PerkinElmer’s exclusive bead-based technology in a sandwich immunoassay format for the detection of phosphorylated proteins in cells. Alpha SureFire assays are a quantitative alternative to Western Blotting, are automation-friendly, miniaturizable, and detect both endogenous and recombinant proteins.

AlphaScreen SureFire

AlphaScreen SureFire has a rich track record of small molecule screening successes:

  • Uses protein A to capture one of the kit’s antibodies
  • 4-hour incubation protocol
  • Need to order the AlphaScreen protein A kit in addition to the AlphaScreen SureFire kit

AlphaLISA SureFire Ultra

The labeling of one of the antibodies with CaptSure, a small peptide tag, is recognized by CaptSure-antibody-coated beads, providing significant improvement over the AlphaScreen SureFire format:

  • Lower background signal, improved sensitivity, and dynamic range
  • No interference from extraneous antibodies; ideal for therapeutic antibody screening
  • AlphaLISA beads are optimal for hemoglobin-containing samples
  • Quicker assay: 2-hour incubation protocol
  • Beads are included in the kit

Alpha SureFire Ultra Multiplex

This format utilizes AlphaPlex technology to read multiple signals from the same well:

  • Improves data quality with internal controls (e.g., total protein) or gathers more data (multiplex phosphorylation events) from a single experiment, resulting in significant time and cost savings
  • Same features as the AlphaLISA SureFire Ultra, beads are included in the kit

LANCE and LANCE (LANthanide Chelate Excitation) Ultra are our TR-FRET (time-resolved fluorescence resonance energy transfer) based, homogeneous (no wash) technologies. One binding partner of interest is labeled with a donor fluorophore (a LANCE Europium chelate) and a second one is labeled with an acceptor fluorophore (ULight™ dye). Upon excitation at 320-340 nm, energy can be transferred from the donor Europium chelate to the acceptor fluorophore if sufficiently close for FRET (~10 nm). This results in the emission of light detectable at 665 nm.

DELFIA

DELFIA (Dissociation-Enhanced Lanthanide Fluorescence Immunoassay) is a proven, robust immunodetection platform based on time-resolved fluorescence (TRF) detection. This detection method, coupled with the unique chemical properties of lanthanide chelates, makes DELFIA a superior alternative to conventional ELISA for research, drug discovery, and clinical applications.

DELFIA offers a combination of high sensitivity, wide dynamic range and long-lasting stability that traditional ELISA cannot match.

Radiometric Detection

Radiometric detection is a direct method that removes the need for target specific antibodies, reducing assay time & variables. A range of isotopes (3H, 14C, 33P, 35S) can be labeled to a variety of without altering biological function to study in the most natural state receptor ligand binding, kinases, G proteins, and other cell signaling pathways.

For high throughput screening, SPA (Scintillation Proximity Assay) technology provides a homogenous radiometric format for a multitude of assays.

  • No separation or washing steps saves time and generates reliable results.
  • A wide range of SPA bead coatings allows a variety of targets to be assayed.
  • A more convenient, cost-effective, and safer alternative to radioactive filter binding assay
  • Amenable to automation for increased throughput.
  • SPA beads can be used in tubes, 96-, 384- or 1536-well microplates and their signal measured on any scintillation counter.
  • Assay miniaturization is possible, reducing total assay volumes and concentrations per well for lower reagents costs and higher throughput.
  • SPA beads are suitable for use with 3H-, 125I-, 33P-, 35S-, and 14C-labeled ligands.
  • No requirement for protein modification.
