Neprilysin (NEP; CD10) is a zinc-dependent metalloprotease responsible for inactivating a variety of peptide hormones (i.e. substance P, glucagon, oxytocin, etc.). NEP is expressed as a 750 amino acid membrane protein but later cleaved to the mature form, a 699 amino acid enzyme. NEP has been shown to be involved in neurodegeneration and tumor development. It degrades and regulates the amyloid beta peptide involved in the development of Alzheimer's disease. In acute lymphocytic leukemia, NEP has been shown to be present on leukemic cells of the pre-B phenotype in 85% of the cases showing promise as a reliable biomarker.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
