DELFIA^® (Dissociation-Enhanced Lanthanide Fluorescent Immunoassay) TRF technology combines the benefits of Time Resolved Fluorescence (TRF) with the use of lanthanide chelates for a highly sensitive, robust and reproducible assay format. With over 20 years of history, and thousands of publications, DEFLIA is ideal for simple to complex matrices and can be used for both biochemical and cell-based assays.

The use of long-lifetime fluorescent lanthanide chelates, rather than other fluorophores, allows for a delay in measurement between excitation and emissions, which occur simultaneously in standard fluorometric assays. By combining the benefits of TRF with the use of lanthanide chelates, DELFIA offers multiple advantages over standard fluorometric detection:

Immunoassays

DELFIA delivers lower detection limits in the femtogram range, sensitivities that are at least 10x greater than standard colorimetric ELISA. DELFIA also offers a broad dynamic range, minimizing the need for time-consuming sample preparation and assay repeats.

Because DELFIA is not an enzyme-based technology, assay performance is not susceptible to degradation of enzyme conjugate activity or substrate signal. Lanthanide fluorescent signals are strong and stable, providing consistent signal measurement even months after an assay has been completed.

Furthermore, DELFIA maintains the wash steps of standard ELISA and so it is ideal when working in complex matrices.

Non-Radioactive Cytotoxicity and Cell Proliferation Assays

DELFIA cytotoxicity and cell proliferation assays provide an alternative to traditional radioactivity-based assays without compromising sensitivity or reliability.

The DELFIA Cytotoxicity assay closely mimics the traditional ⁵¹Cr-release protocol while the DELFIA Cell Proliferation assay couples 5-bromo-2-deoxyuridine (BrdU) incorporation with highly sensitive Europium-based TRF detection.

Binding Assays

DELFIA provides a non-radioactive alternative to measure ligand-binding assays with Europium-labeled ligands, or biotinylated ligands in conjunction with Europium-streptavidin, using whole cells in adherent or filtration formats. It provides the sensitivity of traditional radioactive assays without the risk of exposure. DELFIA also offers a flexible, highly sensitive wash-based assay for protein-protein and other binding interactions that greatly reduces the potential for compound interference.

Enzymatic Assays

Enzymes are important targets in drug discovery. Many marketed drugs today function through inhibition of enzymes mediating disease phenotypes. DELFIA provides a time-resolved fluorescence enzymatic assay compatible with peptide, protein, histone, nucleosome, and other substrates for biochemical assays, or fixed and live cells for cell-based assays.
